Preparation of Houttuyfonate Sodium.

(1) Preparation of 2-tridecanone: First, add 2.3g of sodium metal to the reaction flask, then add 3mL of anhydrous ethanol, heat to 25-30°C, then drop 13.0g of ethyl acetoacetate, 10mL of ethanol and 12.5mL of anhydrous ether into the flask, stir and reflux for 1 hour; then add 21.85g of lauroyl chloride and 30mL of ether to the reaction flask, stir and reflux for 2-3 hours; after cooling, dissolve with dilute sulfuric acid, separate the ether layer, extract the water layer with ether twice, combine the organic layers and wash with distilled water, recover the ether, and obtain 15.7g of the intermediate 2-tridecanone, with a yield of 92%. (2) Preparation of dodecanoyl acetaldehyde: With anhydrous ethanol as solvent, add 1.1g of metallic sodium to the reaction vessel, heat and reflux for 1 hour, dropwise add 8g of 2-tridecanone and 3g of ethyl formate, stir and react for 4-6 hours, let stand overnight, then wash with distilled water, then extract with ether several times, combine the ether solution, dry with anhydrous sodium sulfate and filter, finally separate the ether to obtain 8.3g of dodecanoyl acetaldehyde, with a yield of 90.3%.

(3) Preparation of sodium a-hydroxydodecanoyl ethyl sulfonate: After adding 60ml of saturated sodium bisulfite solution to the reaction vessel, slowly add 6.9g of dodecanoyl acetaldehyde, react at 85-90℃ for 1.5 hours, cool, filter, wash, and dry to obtain 8.1g of sodium a-hydroxydodecanoyl ethyl sulfonate, with a yield of 89.3%. The content was determined by HPLC to be 99.4%.
