Lansoprazole Usage and Function.

Reflux esophagitis, gastric ulcer, duodenal ulcer. Patients with duodenal ulcer take 30mg of this product every day for 2 to 4 weeks, and the effective rate after 4 weeks is 75% to 100%, the healing speed is faster than famotidine and omeprazole, and the pain reduction caused by ulcer is faster than ranitidine. Taking 30mg of this product daily can effectively treat reflux esophagitis, the cure rate after 4 weeks is 63% to 84%, and the cure rate after 8 weeks is 85% to 92%. Action For reflux esophagitis, this product is superior to ranitidine and comparable to omeprazole. After 4 weeks of treatment, it relieved heartburn symptoms significantly better than ranitidine and omeprazole.

This product can be effectively used in the treatment of peptic ulcer or reflux esophagitis patients whose H2 receptor antagonists are not effective. Uses of Lansoprazole.

It can significantly inhibit the secretion of gastric acid caused by basic gastric acid and all stimuli. Lansoprazole is a new generation proton inhibitor, which acts on the last link of gastric acid secretion and strongly inhibits gastric acid, and is used in the treatment of duodenal ulcer and reflux esophagitis. Pharmacological action of Lansoprazole.

Lansoprazole, a substituted benzimidazole derivative, is the second proton pump inhibitor.

In the fine tubule acidic environment of the parietic cells of the stomach, it is converted into an active sulfinamide derivative, which binds to the sulfhydryl group of H+-K+-(ATP) adenosine triphosphatase (H+-K+ -ATPase catalyzes the last step in the gastric acid secretion process), thus passivating H+-K+ -ATPase and inhibiting the central and peripherally regulated gastric acid secretion.

This product is at least as strong as omeprazole in inhibiting gastric acid secretion. In vivo studies in animal models have shown that this product does not inhibit gastric acid secretion as well as H2 receptor antagonists Ranitidine and famotidine, but is as effective as omeprazole. Unlike H2 receptor antagonists, this product inhibits both daytime and nighttime gastric acid secretion, whether taken in the morning or at night. This product can also reduce the amount of gastric acid secretion and inhibit the secretion and activity of pepsin. This product clears the stomach lining of Campylobacter pylori (a bacterium that causes peptic ulcer recurrence).

Product Method of Bulk Lansoprazole Powder.

Add glacial acetic acid and part of 30% hydrogen peroxide to 2, 3-dimethylpyridine, heat and add the remaining hydrogen peroxide. At the end of the reaction, the solvent is steamed under reduced pressure to obtain compound (I). It was slowly added to mixed acid and heated to obtain nitration product (Ⅱ).

The solution of trifluoroethanol reacts for several hours under alkaline conditions, drops the nitration product (Ⅱ), and heats to obtain the substituted product (Ⅲ).

In the substituted product (III), acetic anhydride and concentrated sulfuric acid are added and heated. The solvent was extracted under reduced pressure, and the residue was added to the solution composed of methanol, water and sodium hydroxide, adjusted to alkaline, and reacted at room temperature. Compound (Ⅳ) was obtained.

Compound (Ⅳ) dissolved in chloroform, drops of dichlorosulfoxide, reflux. Steam out the solvent under reduced pressure, add methanol and set aside. In addition, methanol solution of 2-mercaptobenzimidazole and sodium methanol was added, and reflux was carried out. After cooling, the methanol solution for use is dripped and the compound (V) is produced by reflux.

The compound (V) dissolves in methylene chloride and slowly drops into a solution of hydrogen peroxide, vanadium pentoxide and tert-butanol. After the reaction, add 0.5% sodium thiosulfate solution and stir vigorously. Treated with delansoprazole.
