In anesthetized rats, medetomidine (1-100 μg/kg, intravenous) caused a dose-dependent transient decrease in blood pressure and heart rate. In myelospondylotomized rats, medetomidine showed potent vasopressor (PD50 1.7 μg/kg) and sympathoinhibitory (ID50 1.6 μg/kg) effects without affecting basal heart rate.

Medetomidine caused dose-dependent sedation, loss of the forward reflex at high doses (>100 μg/kg), and decreased body temperature. Medetomidine caused a decrease in the turnover of biogenic amines in the brain, a dose-dependent inhibition of norepinephrine (NE) turnover, inhibition of brain dopamine turnover at high doses, and a decrease in serotonin turnover.
