Gentamicin Sulfate Powder is an aminoglycoside antibiotic, a glycoside formed from aminocyclohexylpolyol and aminosugar. 

There are five alkaline centers in gentamicin molecule, which are alkaline compounds, and the drug is the sulfate of gentamicin.

Uses and functions of Gentamicin Sulfate Powder.

Gentamicin Sulfate is a glycoside antibacterial antibiotic linked by an aminosugar and a ligand, it is the sulfate salt of Gentamicin, and its antibacterial spectrum is similar to that of Kanamycin.

It is mainly used for systemic or localized infections caused by gram-negative bacteria such as Escherichia coli, dysentery bacillus, Klebsiella pneumoniae, Proteus mirabilis and Pseudomonas aeruginosa. Ophthalmology is used for conjunctivitis and keratitis. Oral can be used for intestinal infections or colon surgery preparation; can also be used in conjunction with metronidazole or clindamycin to reduce the infection rate of colon surgery.

Gentamicin Sulfate has good antibacterial effect on various gram-negative and gram-positive bacteria, and has good antibacterial effect on various Enterobacteriaceae such as Escherichia coli, Klebsiella, Aspergillus, Salmonella, Shigella, Enterobacteriaceae, Serratiaceae, and Pseudomonas aeruginosa, etc. It has good antibacterial effect on Neisseria and Neisseria influenzae.

Neisseria spp. and Haemophilus influenzae are moderately sensitive to this product. It also has a certain effect on Brucella spp., Plasmodium spp., Fusobacterium spp. and Campylobacter fetus.

It has good antibacterial effect on about 80% of methicillin-sensitive strains of Staphylococcus spp. (including Staphylococcus aureus and coagulase-negative staphylococci), but methicillin-resistant strains are mostly resistant to Gentamicin sulfate.

The effect on Streptococcus spp. and Streptococcus pneumoniae is poor, and Enterococcus spp. are mostly resistant to the product. In recent years, there has been a significant increase in gentamicin-resistant strains of gram-negative bacilli. Synergistic antimicrobial effects can be obtained when the product is used in combination with β-lactams.

Pharmacological Effects of Gentamicin Sulfate Powder.

Gentamicin Sulfate is an aminoglycoside that acts on the ribosomes of bacteria, inhibiting bacterial protein synthesis and disrupting the integrity of the bacterial cell membrane to exert bactericidal effects.

The drug can first pass through the pore protein of the extracellular membrane by passive diffusion, and then enter the cell through the cell membrane by the transporter system, and irreversibly bind to the 30s subunit of the isolated ribosome, leading to the disruption of the A-site, which in turn:

• (1) Preventing the correct localization of aminoacyl tRNA at the A-site, especially hindering the binding of methionyl tRNA, thereby interfering with the assembly of functional ribosomes and inhibiting the formation of the 70s initiation complex.
• (2) Induces a mismatch between tRNA and mRNA codon triplets, causing misreading of the genetic code in the 30s subunit of the intact ribosome, resulting in the insertion of incorrect amino acids into the protein structure, leading to abnormal, non-functional protein synthesis.
• (3) Hinders the binding of termination factors to the A-site, preventing the release of synthesized peptide chains and preventing the dissociation of the 70s intact ribosome.
• (4) Obstructs the process of polyribosome depolymerization and assembly, resulting in ribosome depletion in bacteria.

• 1, Used for the treatment of respiratory tract infections caused by sensitive bacteria, biliary tract infections, intestinal infections, abdominal infections, genitourinary system infections (except simple urinary tract infections), skin and soft tissue infections, burn infections and neonatal sepsis, sepsis.

• 3, Intrathecal injection and coadministration of clindamycin or metronidazole can be used to reduce the incidence of infection after colon surgery.
