Alkaline phosphatase (ALP) is an exoenzyme of osteoblasts, and its expression activity is a distinct feature of osteoblast differentiation. The main physiological function of alkaline phosphatase in the body is to hydrolyze phosphate esters during osteogenesis, providing the necessary phosphoric acid for the deposition of hydroxyapatite, and at the same time hydrolyze pyrophosphate to relieve its inhibitory effect on bone salt formation, which is beneficial to osteogenesis.
Uses and functions of Alkaline.
Alkaline phosphatase has broad specificity for phosphates of alcohols, amines, pyrophosphates, and phenols, and it requires zinc and magnesium or calcium divalent ions for activity. It can be used to dephosphorylate casein and other proteins; it can also be used to dephosphorylate the 5'-end of DNA or RNA to prevent self-ligation.
After dephosphorylation with it, DNA or RNA can also be labeled with radiolabeled phosphates (by T4 polynucleotide kinase). It is used for conjugation with antibodies and other proteins for ELISA, Western blotting, and histochemical detection; it is commonly used to dephosphorylate proteins (such as casein) and nucleic acids. It can be used for protein labeling when high sensitivity is required.
Physiological characteristics of Alkaline.
Alkaline phosphatase is an enzyme that can dephosphorylate the corresponding substrate, that is, it removes the phosphate group on the substrate molecule by hydrolyzing the phosphate monoester and generates phosphate ions and free hydroxyl groups. Such substrates include nucleic acids, proteins, alkaloids, etc. The process of removing the phosphate group is called dephosphorylation or dephosphorylation.
Alkaline phosphatase is a type of phosphatase. The action of phosphatase is opposite to that of kinase. Kinase is a phosphorylase that can use energy molecules such as ATP to add phosphate groups to the corresponding substrate molecules. Alkaline phosphatase has the greatest activity in an alkaline environment. For ALP derived from bacteria, the optimal pH is 8.0, while for ALP derived from cattle, it is 8.5.
