Dinorgestrel is a hybrid progesterone that has the pharmacological advantages of both natural and synthetic progesterone. Dinorgestrel has a high progesterone activity and its oral semi-effective dose is 0.11mg/kg. The endocrine pharmacological properties of dinorgestrel are very desirable, because it only binds to the progesterone receptor, it has no estrogenic, anti-estrogenic, and androgenic activity, and its anti-gonadotropin effect (inhibiting the secretion of follicle-stimulating hormone and lutein hormone) is weak.

A large randomized, double-blind, placebo-controlled clinical trial conducted by Harada et al. showed that after taking 2mg/d for 24 weeks, dinorgestrel was as effective as buserrelin in improving endometriosis related symptoms such as lower abdominal pain, lower back pain and dyspareunia, but the rate of adverse reactions such as vaginal bleeding and decreased bone density was significantly reduced and was better tolerated.
