Production method of Azathioprine Powder:
It is obtained by amination, cyclization, chlorination, nitration and condensation with diethyl oxalate as Raw Material.
1.Amination put diethyl oxalate and methanol into the reaction pot, cool to below 20℃, pass dry methylamine gas to saturation and cool to below 10℃, precipitate crystals, filter and dry, get ethylenediyl dimethylamine, melting point is 210-212℃, yield 95.8%.
2. cyclic combination, chlorination of excess ethylenedicarbonyl dimethylamine and phosphorus pentachloride into the reaction pot in stages, at 70-80 ℃ insulation 2h, placed overnight, reduced pressure evaporation of phosphorus oxychloride, the temperature does not exceed 100 ℃, cooling, adding ice water, stirring, with 30-40% alkali solution to adjust the pH to 9-10, and let stand. Separate out the oily substance, cool the mother liquor, precipitate out the inorganic salt and filter, wash with chloroform and extract the mother liquor with chloroform. Combine the oil and chloroform layers, recover chloroform, distill under reduced pressure, collect the fraction with boiling point 110-115℃ (4.0kPa), and obtain 1-methyl-5-chloroimidazole. The yield is 52%.
3. Nitration of 1-methyl-5-chloroimidazole was added to a glass-lined reaction pot, then nitric acid was added under cooling. Continue to add sulfuric acid dropwise under cooling, after addition, the reaction at 100 ℃ for 2h, and then cooled, adding ice water to precipitate the product, filtered and dried to obtain -5-chloro-1-methyl-4-nitroimidazole. Yield is 86%.
4. Condensation of 6-mercaptopurine, sodium hydroxide, water and 5-chloro-1-methyl-4-nitroimidazole boiled together for 4h, the reaction product was adjusted with acetic acid pH to precipitate azathioprine.
