    The pharmacological effects of theophylline are related to blood concentration. The safety range of its effective blood concentration is very narrow, such as blood concentration of 10-20 μg/ml when the expansion of bronchial tubes, more than 20 μg/ml that can cause toxic reactions.Oral absorption is unstable, its in vivo contouring influencing factors, and individual differences are very large, the concentration in the blood is more difficult to control, so it is easy to occur poisoning.If the dosage is too large, or the speed of intravenous injection is too fast, or the effect of repeated use of the drug is accumulated, there is a possibility of overdose poisoning. And sometimes the symptoms of poisoning are not easy to be found, and even misdiagnosed as the original disease itself; and therefore wrongly further increase the dosage of theophylline, resulting in serious poisoning. Infections can affect theophylline metabolism, increase the concentration in the blood to reach the toxic level. Other factors that can increase theophylline blood concentration are: liver disease, congestive heart failure, respiratory failure, chronic obstructive pulmonary disease, renal failure, metabolic acidosis, hyperthermia, female, young, old and obese. It has been emphasised that intravenous aminophylline in the elderly is prone to toxicity, so it is not advisable to give this drug intravenously to the elderly.
