Production Method of Calcipotriene Powder
Take vitamin D2(Ⅰ) as raw material. Vitamin D2 (12.5g, 31.5mmo1) was dissolved in 50ml liquid sulfur dioxide, stirred and refluxed for 30min, evaporated the sulfur dioxide, and the residual solution was dried under vacuum. Then dissolved in 100ml dimethylformamide, then added imidazole (4.5g, 66mmo1) and tert-butyldimethylsilyl chloride (5.0g, 33mmo1), stirred at 20°C for 90min. The reaction solution was partitioned between 500ml ethyl acetate and 200ml water.
The organic layer was partitioned, washed with water and brine, dried, and concentrated. The residue was separated by chromatography (eluted with petroleum ether solution of 30% ether) to give 9.4 g of compound (IIa), 52% yield, melting point 117-118°C (decomposition); and 7.7 g of compound (IIb), 42% yield, melting point 121-122°C (decomposition).
