    Theaflavine powder Usage and Synthesis.

    The purified theaflavins are orange-yellow needle-shaped crystals with a melting point of 237-240°C. They are easily soluble in water, methanol, ethanol, acetone, n-butanol and ethyl acetate, but poorly soluble in ether. They are insoluble in chloroform and benzene.

    The theaflavins solution is bright orange-yellow, and the aqueous solution is weakly acidic with a pH of about 5.7. The color is not affected by the pH of the tea extract, but it has a tendency to auto-oxidize in alkaline solutions, and this tendency increases with increasing pH.

    Preparation of Theaflavin powder.

    Weigh 100g of tea leaves, add 1000ml of deionized water (90℃) and extract for 30min, take out the extract; add 500ml of deionized water (90℃) and extract for 30min; combine the extracts, filter through 300 mesh filter cloth, and treat with 0.05um ceramic membrane to obtain theaflavin microfiltrate.

    The theaflavin microfiltrate was pumped into a two-stage series centrifugal extractor from the heavy phase inlet, and 1/2 of the volume of the theaflavin microfiltrate ethyl acetate solution was pumped into the two-stage series centrifugal extractor at the same time. The speed of the two series centrifugal extractors was 3000rpm. The light phase outlet liquid (i.e., ester phase solution) was collected, and the theaflavin extract was obtained by vacuum concentration at 40℃ and spray drying. The purity of the theaflavin was 21.35%.

    Pharmacological Action of Theaflavin powder.

    Theaflavins can inhibit the proliferation of human nasopharyngeal carcinoma CNE2 cells and induce their apoptosis. Its effect is time- and concentration-dependent, and its mechanism of action may be related to regulating the expression of cell proliferation and apoptosis genes.

    Theaflavins can reduce serum uric acid in hyperuricemic mice. Its effect of reducing serum uric acid is related to inhibiting the activity of xanthine oxidase and regulating the mRNA and protein expression levels of related anion transporters.

    Theaflavins can enhance the sensitivity of cervical cancer cells to cisplatin, and its mechanism may be related to increasing the expression of apoptosis-related proteins CleavedCaspase3, CleavedCaspase9, and PARP and inhibiting the expression of NF-κB signaling pathway-related proteins p-IκBα, p-NF-κB, and p-Akt.

    Theaflavins can effectively delay the inflammatory degeneration process of rat chondrocytes by enhancing the activity of chondrocyte synthesis factors, weakening the activity of decomposition factors, and inhibiting cell inflammatory response, and have a certain protective effect on inflammatory chondrocytes.
