Preparation of Raspberry Ketone.
Using p-hydroxybenzaldehyde and acetone as raw materials
The 4-p-hydroxyphenyl-3-butene-2-ketone was synthesized by Claisen-Schmidt condensation under base catalysis and optimized by orthogonal experiment (88.4% yield). Raspberry ketone was synthesized by hydrogenation under palladium-carbon catalysis (84.0% yield). P-hydroxyphenyl-3-butene-2-ketone was synthesized with NaOH solution as Claisen-Schmidt condensation catalyst. The yields of raspberry ketone were 72.55% and 65.7% using 5% Pd /C and nickel formate as the second step catalysts, respectively.
Orthogonal test was used to study the condensation reaction conditions of p-hydroxybenzaldehyde and acetone, and the optimal process conditions were as follows: The ratio of n(p-hydroxybenzaldehyde)∶n(acetone)∶n(NaOH) = 1.0∶ 25∶ 25(reaction at 30 ℃ for 5 h) was followed by conventional hydrogenation reduction, vacuum distillation and mixed solvent recrystallization to obtain raspberry ketone (yield 61.8%). Claisen-Schmid condensation reaction was catalyzed by NaOH, nickel formate as catalyst and isopropyl alcohol as solvent to produce raspberry ketone with yield of 42.0%.

Hydrogenation route
First, 3.5mol 15% sodium hydroxide was added to the reactor, and 2mol acetone and 1mol para-hydroxybenzaldehyde were added to the mixture under stirring, and the temperature was controlled at 20-30 ℃. After the drip, the reaction was continued to stir for 20h. The solid sodium salt of 4-p-hydroxyphenylbutene-3-keto-2 was obtained by filtration, dissolved with water, then acidified with hydrochloric acid to precipitate 4-p-hydroxyphenylbutene-3-keto-2, and dried after filtration for use.
Add the above intermediate 100g and nickel catalyst 5g in the autoclave, replace the air with hydrogen, and heat it to 120 ~ 140℃, and react at 0.98MPa pressure for 1.5h. After cooling, the catalyst was filtered, the content of raspberry ketone was 89%, and the hydrogenation yield was 92%. The crude product is distilled once under vacuum and recrystallized 1 ~ 2 times to obtain white crystal with a melting point of 82℃.
