Hydroxychloroquine sulfate is an anti-malarial drug with anti-inflammatory, immunomodulatory and anticoagulant effects. At present, hydroxychloroquine is mainly used in the clinical treatment of rheumatoid arthritis, juvenile chronic arthritis, discoid lupus erythematosus and systemic lupus erythematosus, but the clinical effect is slow, often taking 2 to 4 months. The pharmacological action of this drug is not very clear, it may be related to the stabilization of lysosomal membrane, inhibition of prostaglandin formation, inhibition of chemotaxis of polymorphonuclear cells and phagocytosis, may interfere with the formation of monocyte interleukin-1 and inhibit the release of neutrophil superoxide.

Hydroxychloroquine sulfate has similar pharmacological action, pharmacokinetics and metabolic process as chloroquine. After oral administration, hydroxychloroquine is rapidly and almost completely absorbed. In one study, after a single dose of 400mg of hydroxychloroquine was given to healthy volunteers, the average plasma peak concentration was in the range of 53-208ng/ml, with an average level of 105ng/ml. The mean time for plasma to reach peak concentration was 1.83 hours. Depending on the time after administration, the mean plasma elimination half-life changes as follows: 5.9 hours, 26.1 hours, and 299 hours, respectively, at -10, 10-48, and 48-504 hours after peak plasma concentration. Parent compounds and metabolites are widely distributed in the body and are eliminated primarily through urine. In one study, 3% of the dose administered was observed over 24 hours.

Hydroxychloroquine sulfate has a cumulative effect and takes several weeks to exert its beneficial effects, while minor adverse reactions may occur relatively early.
If rheumatic diseases do not improve after 6 months of treatment, treatment should be terminated. In the case of light-sensitive diseases, treatment should only be given with maximum exposure to daylight.
④ All patients should undergo ophthalmology examination before starting treatment with this product. Contraindicated in patients with pre-existing macular disease of the eye.
