preparation of Heparin Lithium.
1) Crude heparin enzymolysis: dissolve crude heparin sodium, adjust the PH value to 7.5‑9.0, temperature to 50‑55℃, add heparin enzymolysis pancreas enzyme, heat for 4‑5 hours, adjust the PH value 9.0‑11, temperature rise to 85-90℃, cool, filter to remove the sediment, filtrate with ethanol precipitation, collect the sediment (A);
2) Deprotein: Step 1 The resulting precipitate (A) is dissolved in water, stirred, adjusted PH to 9-11, added protein flocculating precipitator, stirred, left, filtered to remove precipitate, collected filtrate (D), adjusted PH to 11-13, stirred, left, filtered to remove precipitate, collected filtrate (E), adjusted PH to neutral, precipitated with ethanol, Collection of sediment (B);
3) Oxidation and decolorization: the sediment obtained in step 2 (B) is dissolved in water, oxidized with hydrogen peroxide for 24‑48 hours, adjusted PH to 10.5‑11.5, filtered to remove the sediment, collect the filtrate (F), adjust the PH value of the filtrate (F) to 6.5‑7.5, precipitate with ethanol, collect the sediment (C);
4) Resin exchange: Dissolve the precipitate (C) obtained in step 3 with water, add LiCl, stir to dissolve, carry out ion exchange through the resin column, collect the effluent from the column, and obtain heparin lithium solution;
5) Precipitation, dehydration and drying: the lithium heparin solution obtained in step 4 is filtered, precipitated with ethanol, the sediment (G) is collected, the sediment (G) is dehydrated, and then vacuum dried and crushed to obtain the lithium heparin fine.
