    L-malic acid contains natural emollient ingredients, which can remove wrinkles on the surface of the skin and make the skin white, smooth and elastic, so it is favored in cosmetic formulations; L-malic acid can be formulated with a variety of flavors and fragrances, and is used in a variety of daily-use chemicals, such as toothpaste, shampoo, etc.; it is used in foreign countries as a new type of detergent auxiliary in place of citric acid, and is used in the synthesis of high-grade special detergents.

    Uses and Function of L-Malic Acid.

    Used in the manufacture of esters; used as complexing agent and flavoring agent. It can be used in all kinds of food according to GB2760-90 in China. As an acidifier, it can be used instead of citric acid (about 80%), especially suitable for jelly and fruit-based food.

    It has the function of keeping the color of natural fruits, and it can also be used as the extraction auxiliary of pectin, the agent for promoting the growth of yeast, the agent for preparing salt-free soy sauce and vinegar, and the emulsifying stabilizer for improving the taste of pickles and margarine, mayonnaise, etc. It is widely used in all kinds of preservatives, flavoring agents.

    Preparation of L-Malic Acid.

    (1) Preparation of L-malate and fumarate mixture according to the cell transformation method (DE3310849) in the prior art. Aspergilluswentii strain isolated from soil was cultured in a culture solution of sugar 30 g/L, diammonium hydrogenphosphate 3 g/L, ammonium sulfate 2 g/L, potassium chloride 2 g/L, magnesium sulfate 1 g/L, and sodium fumarate 5 g/L for 48 h. The cells were isolated by filtration.

    A 2% carrageenan solution was prepared at 60-70°C, and the isolated cells were added at 10% (w/w) when the temperature was lowered to 50°C. The cells were stirred well and poured into a dish to solidify to make 5mm×5mm×5mm immobilized cells. Then it was loaded into an 8 cm × 60 cm glass column.

    A 1.4 M sodium fumarate solution was flowed through the immobilized cell column at a rate of 1000 ml/h to obtain a mixed solution of L-malate and fumarate.

    (2) 1000 ml of the mixed solution obtained by step (1) was taken, containing 186 g of sodium L-malate and 56 g of sodium fumarate. the solution was heated to 60° C., 170 g (molar ratio 1.05:1) of fumaric acid was added with stirring, and the reaction was carried out with stirring for 3.5 h. The reaction was carried out at pH = 3.2, cooled to 15-20° C., filtered, and the filter cake was washed with a small amount of water and then neutralized with NaOH to pH = 7-8 for cell transformation.

    The resulting filtrate was 980 ml, containing 40 g of monosodium fumarate. heated to 85 ℃, passed through 1500 ml 001x7 strongly acidic cation exchange column (diameter 6 cm, height 60 cm), collected the eluent was concentrated under reduced pressure, the concentration of L-malic acid was increased to 40%, cooled to 10-15 ℃, and filtered to recover fumaric acid.

    The filtrate was further concentrated under reduced pressure to L-malic acid concentration to 80~82%, cooled to 15°C, 91 g of L-malic acid was crystallized, and the mother liquor contained 46 g of L-malic acid, and the mother liquor could be recycled.

    The yield of L-malic acid was equivalent to 97%.

    Product Methods of  Bulk L-Malic Acid Powder.

    Malic acid is widely found in nature, is the product of animal and plant metabolism, and is the main acid component in apples. L-malic acid can be obtained by boiling the juice of unripe apples, grapes, and peaches, adding limewater, generating a calcium salt precipitate, which is then transformed into a lead salt, and then treated to generate free acid.

    Fermentation method is also possible to realize industrialization, using mutated yeast species for fermentation, which can obtain L-malic acid-based special bacteria for acid generation, or using special bacteria that mainly produce malic acid for fermentation, which can obtain L-malic acid, but the process is more complicated than the synthesis method, and the cost is higher. In Japan, L-malic acid is produced by biosynthesis.

    First, Breridacterium ammon-agenescell was immobilized with polyacrylamide. A suspension of cells in saline was mixed with acrylamide and appropriate cross-linking agents and polymerization promoters, and the resulting gel was made into 3-mm-diameter particles. After washing, they were immersed in sodium transbutylene succinate containing about 0.3 bovine bile, and the solution was maintained at 37°C for 20 h. Cells treated in this way were used as packing material for the reaction columns.

    1 L of sodium transbutylene succinate containing 1 mol was passed through the reaction column at 37°C, and malic acid was separated from the effluent by the usual method to produce a pharmaceutical grade product of L-malic acid from transbutylene succinate in about 70% yield.
