Shikonin Powder Usage and Synthesis.
Shikonin is a naphthoquinone compound derived from lithospermum, which has anti-inflammatory, antibacterial, antiviral and anti-tumor effects. Shikonin and its derivatives can be produced by large-scale plant cell culture technology.
Shikonin has also been found to inhibit DNA damage caused by chemotherapy drugs. In the treatment of periodontitis, shikonin is widely used due to its anti-inflammatory and low toxicity. These properties of shikonin give it broad application prospects in the medical field.
Uses of Shikonin.
Edible purple pigment. Used for coloring spicy meat and poultry cans, dosage 0.35-0.56g/kg. Put the crushed lithospermum into a fine sieve, pour 160-170℃ hot oil on it, so that the pigment dissolves in the oil. The oil flows out through the fine sieve, mixes with the chili powder in the container, and after fully stirring, boils into a mixed sauce for later use.
Pharmacological effect of Shikonin.
(1) Hypoglycemic effect Lithospermum leaf extract and lithospermum polysaccharide (A, B, C) have obvious hypoglycemic effect.
(2) Antibacterial effect Lithospermum has an inhibitory effect on Jingke 68-1 virus in vitro and Staphylococcus aureus. The anti-parainfluenza virus effect of L-shikonin was studied by hemagglutination reaction and cytopathic effect assay. The results showed that its toxicity was low within the concentration range used in the experiment, and it had certain in vitro anti-influenza virus activity and direct killing effect on parainfluenza virus.
(3) Effect on blood coagulation Intraperitoneal injection of lithospermum pigment components (shikonin, acetylshikonin) did not affect blood coagulation time, but could prevent the anticoagulant effect of heparin.
(4) Anti-tumor effect Lithospermum extract has a certain inhibitory effect on the late DNA synthesis (G2 phase) of Hela cells.
(5) Anti-cancer effect: Shikonin inhibits proliferation, promotes apoptosis and induces cell cycle arrest in human choriocarcinoma resistant cell lines (JAR/MTX) cultured in vitro. Experiments have shown that the inhibition rate of shikonin on the growth of choriocarcinoma resistant cells increased significantly with the increase of dose and the extension of action time.
Product Method of Bulk Shikonin Powder.
The root of Lithospermum officinale was used as raw material, extracted with 4 times the amount (mass) of refined soybean oil at 165-170°C for 6 minutes, and then the extract was extracted twice with 95% ethanol (volume ratio) slightly more than the soybean oil; the ethanol extract was concentrated under a vacuum of 80 kPa, and then the insoluble matter was filtered out to obtain the finished product.
