    The seed media for the fermentation culture of polycidin polyspora were: soybean hydrolysate 3%, yeast paste 0.3%, MgSO4·H2O 0.2%, glucose 0.5%, maltose.4%, deionized water 1L, sterilized at 121℃ for 30min. The dose is 10%. In large-scale production, the preferred carbon sources are glucose and maltose, but also ribose, xylose, fructose, galactose, mannose, mannitol, soluble starch, potato dextrin, methyl oleate, oil, etc. The preferred nitrogen is raffinose, peptonized milk and digested soybean powder. Fish meal, corn infusion, yeast paste, hydrolyzed casein and beef paste can also be used. During industrial fermentation, the dissolved oxygen in the tank should be maintained at more than 60%, preferably more than 65%, by ventilation and stirring speed. The pressure inside the tank is 0.034MPa.

    After fermentation for 7 ~ 10 days, the same volume of acetone was added, the bacteria were removed by filtration, the pH value of the filtrate was adjusted to 13, and the resin was adsorbed. Polycidin A and D were gradient eluted with a solution of 0 ~ 95% methanol: acetonitrilene = 1:1 (containing 0.1% sodium acetate), and the eluents were collected in stages to obtain polycidin eluents, which were concentrated to obtain polycidin concentrate. The concentrated solution was diluted with petroleum ether, on A silica gel column, gradient elution with petroleum ether and methanol, and then collected in stages to obtain dicidin A and D elutes.
