The principle of urinary N-acetyl β-D aminoglucosidase is to release free p-nitrophenol under the action of N-acetyl β-D aminoglucosidase (NAG), which has a strong absorption peak at 405 nm, and the amount of p-nitrophenol produced can be measured to calculate the NAG activity.
