Ceramides can be synthesized by three different pathways, namely de novo synthesis pathway, sphingomyelase pathway and remedial pathway. De novo synthesis pathway is the formation of 3-ketosphinganine by serine palmitoyl CoA catalyzed by serine palmitoyl transferase on the cytoplasmic surface of the endoplasmic reticulum (ER), and then dihydrosphinganine. It is then acylated by ceramide synthase to produce dihydrosphingosine, and finally dehydrogenated to form ceramide. The remedy is that glycosylated sphingolipid and sphingomyelin form sphingosine under the action of lysosomal glycosidase, acid sphingosphingolase and acid ceramidase, and isodihydrosphingosine can be acylated to form ceramide in the ER by ceramide synthetase. In addition, ceramides can be produced by hydrolysis of sphingomyelinase (SMase) on the cell membrane.
