It also blocks dopamine D2 receptors in the substantia nigra-striatal pathway to produce extrapyramidal responses, and blocks dopamine D2 receptors in the nodal-funnel pathway to cause endocrine disruption.

Chlorpromazine also antagonizes adrenergic alpha receptors and M-cholinergic receptors, thus its wide range of pharmacological effects underlies its long-term application leading to serious adverse effects.
