Iodixanol powder Usage and Synthesis.

Iodixanol, chemical name 5,5'‑((2‑ hydroxyl ‑1,3‑ propane) bis (acetylimide) bis (N,N ‑2, 3‑ hydroxy propyl) bis (N,N ‑2, 3‑ hydroxy propyl)‑2,4,6‑ triiodol ‑1,3‑ phenylphthalamide, developed by Nycomed, Norway, It is a new type of non-ionic X-ray contrast agent (290mosmol/kg osmotic pressure) and is the only contrast agent that is isotonic with plasma when applied intravascular.

Uses of Iodixanol.

X-ray contrast medium. Angiocardiography, cerebrovascular angiography (routine with i.a. DSA), peripheral arteriography (routine with i.a. DSA), abdominal angiography (i.a. DSA), urography, venography, and CT-enhanced examination are performed in adults.

Preparation of Iodixanol.

Combine potassium hydroxide (112.7g,2mol), the intermediate "ioxanol?" 2 "(1kg,1.33mol) dissolved in water (1.25L). Potassium dihydrogen phosphate (45.5g) and dipotassium hydrogen phosphate (106.9g) were added and stirred for a while, and epichlorohydrin (68.1g,0.8mol) was added. After stirring for 48h, the HPLC yield of ioxanol was 92%. Concentrated hydrochloric acid was added to the reaction system to adjust the PH value of the solution to 7, 3Kg strong acidic ion exchange resin was added and stirred for 1 hour, then filtered and washed, 3kg strong basic ions were added to the mother liquor, and the resin was changed and stirred for 1 hour, then filtered and washed, and the mother liquor was reduced pressure and concentrated to obtain white solid ioxanol crude. The crude product was recrystallized with a mixture of methanol and water to obtain a white solid quality of 782 g iodoxanol (liquid purity of 99.6%).
