Trimethoprim / TMP is a lipophilic and weakly basic ethylaminopyrimidine bacteriostat, also known as sulfonamide synergist, methoxybenzylaminopyrimidine, antimicrobial synergist, methoxybenzylaminopyrimidine, methoxybenzylpyrimidine, trimethoprim, trimethoprim, a white or off-white crystalline powder at room temperature, odourless and bitter in taste, slightly soluble in chloroform, slightly soluble in ethanol or acetone, virtually insoluble in water, soluble in Slightly soluble in chloroform, slightly soluble in ethanol or acetone, almost insoluble in water, soluble in glacial acetic acid.
The antibacterial spectrum of Trimethoprim/TMP is similar to that of sulfonamides, but the antibacterial effect is stronger, it is effective against Escherichia coli, Proteus mirabilis, Pneumocystis pneumoniae, Staphylococcus saprophyticus, and many kinds of gram-positive and gram-negative bacteria, but it is ineffective against Pseudomonas aeruginosa infections, the lowest inhibitory concentration is often lower than 10mg/L, and it is easy to cause bacterial resistance when used singly, therefore it is not usually used alone, it is mainly used in the form of a combination with sulfonamide. It is mainly used in combination with sulfonamides to treat urinary tract infections, intestinal infections, respiratory infections, bacillary dysentery, enteritis, typhoid fever, meningitis, otitis media, otorhinitis, sepsis and soft tissue infections, etc. It is effective against typhoid fever and paratyphoid fever. The antibacterial principle of Trimethoprim / TMP is to interfere with bacterial folate metabolism. The main mechanism of action is to selectively inhibit the activity of bacterial dihydrofolate reductase, so that dihydrofolate can not be reduced to tetrahydrofolate, and the synthesis of folic acid is the main component of nucleic acid biosynthesis, so the product prevents the synthesis of bacterial nucleic acids and proteins, and methocarbamol (TMP) and the bacterial enzyme of dihydrofolate reductase binds to the enzyme of mammals than the binding of 50,000 ~ 60,000 times more closely. Combined with sulfonamides can make the bacterial folic acid synthesis and metabolism was doubly blocked, there is a synergistic effect, so that the antibacterial activity of sulfonamides enhanced, and can make the bacteriostatic effect into bactericidal effect, reduce drug-resistant strains.
