Naproxen sodium is a kind of non-steroidal anti-inflammatory and analgesic drug that is easily tolerated by people. It belongs to phenylpropionic acid compound, which plays an anti-inflammatory and analgesic effect by inhibiting the synthesis of prostaglandins. After oral administration, naproxen sodium is absorbed rapidly, and the plasma concentration reaches its peak within 2 to 4 hours. Food has little effect on absorption rate, naproxen is highly bound to plasma protein (99%), and plasma concentrations range from 23 to 40mcg/ml. Metabolized to demethylate and then combined with glucuronide, elimination half-life of 12 to 15 hours, mainly excreted by urine. Naproxen sodium Powder Usage and Synthesis.
Naproxen sodium is suitable for rheumatoid arthritis, osteoarthritis, ankylosing spondylitis, Juvenile rheumatoid arthritis (Juvenile arthritis), tendinitis, bursitis, and acute gouty arthritis. It relieves symptoms of arthritis pain, swelling, and limited mobility. Psoriatic arthritis and Reiter's syndrome can also be treated with this product. Symptom relief was similar to ibuprofen, fenoprofen, aspirin, sulinac, and indomethacin, but the incidence and severity of gastrointestinal and neurological adverse reactions were lower. In addition, this product can also inhibit platelet aggregation and prolong bleeding time like aspirin, but it is reversible and can be recovered after withdrawal.
Uses of Naproxen sodium.
Naproxen sodium has a strong anti-inflammatory and analgesic effect, and its main biological effects are:
(1) Inhibiting the activity of prostaglandin synthetase, thereby reducing capillary permeability;
(2) Reduce the production of peroxides by macrophages and play an anti-inflammatory role;
(3) Stimulating T inhibitory cell activity;
(4) Reduce pain sensitivity. Symptom relief was similar to ibuprofen, fenoprofen, aspirin, sulinac, and indomethacin, but the incidence and severity of gastrointestinal and neurological adverse reactions were lower.
In vitro study of Naproxen sodium.
Naproxen acted on intact cells and inhibited COX-1 and COX-2 with an IC50 of 2.2 μg/mL and 1.3 μg/mL, respectively. In vitro, Naproxen decreased LPS-induced production of PGE2 and TX2B in rats and humans, with IC50 values of 30.7 μM and 79.5 μM, respectively, when inhibiting PGE2 and 72.4 μM and 48.3 μM, respectively, when inhibiting TX2B. Naproxen inhibited human platelet TX2B production and LPS-induced human monocyte TX2B production with IC50 of 5.7 and 6.4, respectively, and slightly selectively inhibited constitutive and inducible COX-2, with a COX-1 to COX-2 IC50 ratio of 6.3. Only high concentration Naproxen could induce apoptosis of HCA-7 colon cancer cells for 48 hours, IC50 was 1.45 mM.
