Tazarotene is A third generation retinoic acid dermatologic drug, which is a precursor of retinoid for skin external use. Tazarotene can regulate the differentiation and proliferation of epidermal cells, and is mainly suitable for the treatment of plaque psoriasis vulgaris. Tazarotene cream can also be used to treat facial acne. For clinical use, it contains 15mg/30g of the active ingredient of this product. Tazarotene Usage and Synthesis.
tazarotene gel, a topical formic acid preparation, can be used to treat psoriasis. Early studies have shown that topical application twice daily for eight weeks in approximately 50% of patients with mild to moderate psoriasis results in relief of at least 75% of skin lesions. However, the total elimination of plaque was less than 10%. tazarotene gel is available in 0.05% and 0.1% concentrations. 0.1% gel for topical use once a day and twice a day makes no difference. However, 0.5% gel is less effective when used externally once a day.
Tazarotene is a synthetic ethylene retinal, which is rapidly converted to tazarotene acid after topical administration and exerts pharmacological effects. The binding of tazarotenic acid to retinuronic acid receptor gamma (RAR-γ) in the skin may improve clinical symptoms by restoring normal skin differentiation and reducing skin inflammation.
Uses of Tazarotene.

In HL-60 human myeloid leukemia cells, Tazarotene causes ERK activation, RB tumor suppressor protein phosphorylation, G0 block, and myeloid differentiation. Tazarotene is able to drive the early or late part of the period leading to differentiation or G0 arrest, and is interchangeable with the Raralpha-selective ligand. By interacting with specific nuclear retinoic acid receptors (RARs), Tazarotene therapeutically regulates gene transcription, thereby regulating three important causative factors in psoriasis. Tazarotene inhibits proliferation of fibroblasts and synthesis of DNA and collagen. Tazarotene down-regulates markers of keratinocyte differentiation, keratinocyte proliferation, and inflammation. tazarotene also upregulates three new genes, TIG-1 (Tazarotene-inducible gene-1), TIG-2, and TIG-3, which may mediate anti-proliferative effects. In retinoid responsive breast cancer cell lines, Tazarotene causes growth inhibition by upregulation of TIG3.
