Obeticholic acid is the first drug developed in 20 years for the treatment of cholestatic liver disease in patients who do not respond adequately to ursodeoxycholic acid or who cannot tolerate it.
Uses and functions of Cyanoacetic Acid Powder
PBC is a disease caused by chronic cholestasis due to bile duct dysplasia or bile acid metabolism disorders, which results in lipolysis of cell membranes, altered permeability, and efflux of cellular contents, and ultimately progresses to hepatic fibrosis and cirrhosis.
Currently, ursodeoxycholic acid is the main drug used in the clinical treatment of PBC, which mainly stimulates hepatobiliary secretion through the intracellular signalling molecule mitogen-activated protein kinase (MAPK) pathway, and protects epithelial cells from bile acid damage.
Obeticholic acid is a human bile acid mimetic, a semi-synthetic goose deoxycholic acid, and a specific agonist of the farnesyl ester derivative X receptor.
Developed for the treatment of primary biliary cirrhosis PBC, non-alcoholic steatohepatitis and other liver and intestinal disorders, it has been demonstrated in animal studies to improve insulin resistance and reduce liver fat content.
Pharmacological Effects of Cyanoacetic Acid Raw Powder
- The synthesis of obeticholic acid started from cheap and readily available goose deoxycholic acid in CHCl3/Cl2CH2 medium, 7α-OH of goose deoxycholic acid was selectively oxidised to 7-carbonyl by PCC reagent at room temperature to produce compound 1;
- In the same medium CHCl3/Cl2CH2, compound 1 was reacted with 3,4-dihydro-2H-pyran in the presence of catalyst p-toluenesulfonic acid (p-TsOH) to form compound 2;
- Compound 2 was alkylated by ethyl iodide (Et-I) in the presence of n-butyl lithium (n-BuLi) and the enolated structure LDA/HMPA to yield 6-ethyl, and then pyridinium p-toluenesulfonate (PPTS) to produce compound 3;
