    Alitame is a dipeptide synthetic sweetener synthesized from aspartic acid and alanine, its sweetness is more than 2,000 times that of sucrose, 10 times higher than aspartame, with the advantages of good taste, high sweetness and low calorie, it's one of the substitutes of sucrose, with stable nature, it's widely used in the food industry, and it can be added to dairy products, frozen drinks, preserves, beverages, gelatin-based confectionery and jellies. GB2760-2014 "Food Safety National Standard for the Use of Food Additives" has made clear provisions for the maximum use limit of the above foods. As a strong sweetener, Allysine is generally regarded as a non-toxic and stimulating substance, and the WHO stipulates that the allowable daily intake of Allysine can be up to 0.1mg/kg.

    Alitame Usage and Synthesis.

    Uses of Alitame.

    It is a new type and high sweetness sweetener. Our country stipulates that it can be used in gummies, Chenpi, plums, plums and dried prunes, with a maximum usage of 0.3g/kg; in beverages, ice creams and sorbets, with a maximum usage of 0.1g/kg; and it can also be used as tabletop sweeteners, with a maximum dosage of 0.015g/packet and slice.

    Preparation of Alitame.

    A method for the preparation of an alitame standard substance, comprising each of the following steps:

    (1) Preparation of alli sweet solution: the raw material for the preparation of alli sweet standard substance is industrial grade alli sweet, the raw material of alli sweet is dissolved in water and dissolved by ultrasonication, and then filtered by 0.22 μm filtration membrane, to obtain alli sweet solution;

    (2) Liquid chromatography preparation: the alitame solution was injected into the preparative chromatographic separation of pure fractions, the chromatographic conditions for the column for PREP-ODS (H)-KIT (250 mm × 20 mm, 5 μm); mobile phase: 40% (v/v) methanol aqueous solution; flow rate: 10 mL/min; column temperature: 30 ℃; running time: 15 min; detection wavelength: 210 nm.

    The injection volume was 0.2mL, and the fractions within 7.5-9min were collected;

    (3) Fraction collection: the fraction was received in a brown bottle with the outer wall wrapped with tin foil filled with nitrogen gas beforehand, and then a small amount of hydrochloric acid was added to neutralize to pH7;

    (4) drying: the fraction is placed in a rotary evaporator using decompression distillation method at 55 ℃ -75 ℃, and then placed in a lyophilizer -50 ℃ in the freeze-drying for 24 hours, freeze-dried aleutic acid solid grinding and crushing, and then again freeze-dried for 24 hours has been as far as possible to remove the free water, get aleutic acid standard material; after determination, the purity of the standard material is 99.4%.

    (5) Preservation: in a relatively independent and clean glove box on the preparation of pure alli sweet solid using 2mL brown sample bottles for dispensing, with PTFE-lined black cap sealing, and the use of plastic bags independent vacuum plastic sealing.
