Product Overview:
Valsartan is a specific angiotensin II (AT1) receptor antagonist and another non-skin AT1 receptor antagonist used in clinical practice after chlorosartan, which plays a key role in the regulation of systemic blood pressure and the maintenance of electrolyte-fluid balance. It selectively acts on the AT1 receptor subtype, blocking the binding of Ang II to the AT1 receptor (its specific antagonism of the AT1 receptor is about 20,000 times greater than that of the AT2 receptor), thereby inhibiting vasoconstriction and aldosterone release and producing a hypotensive effect, but not inhibiting aldosterone release due to potassium ions (K+).

Valsartan Powder Usage and Synthesis.
Valsartan, an angiotensin receptor antagonist, can be used for all types of hypertension and has a good protective effect on the heart, brain and kidney. It can be used routinely in hypertensive patients with myocardial infarction, heart failure, proteinuria, diabetes mellitus, etc., and can be used in combination with diuretics (such as hydrochlorothiazide).
Uses and functions of Valsartan.
Valsartan is a widely used clinical anti-hypertensive drug with the advantages of few side effects and good tolerability, and it can also be used for the treatment of hypertension in patients with diabetes mellitus and nephropathy.

Drug interactions of Valsartan.
1. No clinically significant drug interactions have been identified. The following drugs have been studied: cimetidine, warfarin, furosemide, digoxin, atenolol, indomethacin, hydrochlorothiazide, amlodipine and glibenclamide.
2, Because valsartan is hardly metabolised, no clinical interaction with drugs that induce or inhibit the cytochrome P450 system has been found.
3, Although valsartan is mostly bound to plasma proteins, in vitro experiments have not found it to interact with other plasma protein-binding drugs (e.g. diclofenac, furosemide, warfarin) at this level.
4, When combined with potassium-preserving diuretics (e.g. spironolactone, amphotericin, amiloride), potassium supplementation or the use of potassium-containing preparations may lead to an increase in blood potassium concentration. Therefore, care is needed when using the drug in combination.

2'-Cyanobiphenyl-4-aldehyde (I) and L-valine methyl ester were subjected to reductive amination and the compound (II) obtained was then acylated with valeroyl chloride and chromatographed to give compound (II). Then it was reacted with Bu3SnN3 to introduce tetrazole and then hydrolysed to give the product.
