The drug was approved for marketing by the US FDA in 2001 for the treatment of central mammalian diseases caused by coccidia such as Neosporahughsi (Hong) and Sarcocystis (Sarcocystis). Compared with the previous anticoccidial drug toltrazuril, the most important feature of patrozuril is that it can cross the blood-brain barrier and directly kill the cerebral and cerebrospinal fluid located in the brain and the protozoa, effectively relieving the symptoms of EPM.
Research shows that Patrovaleri kills coccidia in animals without affecting the production of autoimmunity,and with the enhancement of autoimmunity, sick animals can gradually get rid of the dependence on drugs,while research shows that Patrovaleri has a lower level of toxic effect than Toltrazuril.
Therefore, Patrozulin has better prospects for market development and application.
Uses of Ponazuril.
It is used for the treatment of central mammalian diseases caused by coccidia such as Neosporahughsi (Hong) and Sarcocystis (Sarcocystis), etc. Compared with the previous anti-coccidial drug Totrizolide, the most important feature of Patrozolide is that it can pass through the blood-brain barrier and kill the protozoa located in the brain and the cerebrospinal fluid directly to effectively alleviate the symptoms of EPM.
Pharmacological Effect of Ponazuril.
(1) Pharmacodynamics
Toltrazuril is a triazinone compound with broad-spectrum anticoccidial activity. It is widely used in chicken coccidiosis.
The site of action of toltrazuril on coccidia is very wide, and it has effects on both asexual cycles of coccidia,such as inhibition of schizonts, nuclear division of small gametophytes and wall-forming bodies of small gametophytes.
As the product interferes with the nuclear division and mitochondria of the coccidian cells, it affects the respiration and metabolic function of the worm, plus it can make the endoplasmic reticulum of the cells expand, and serious vacuolation occurs, thus it has the effect of killing coccidia.
(2)Pharmacokinetics
Poultry internal dose of toltrazuril, about more than 50% of the drug absorption, after absorption of the drug is mainly pooled in the liver, kidney, but rapidly metabolised into sulfone compounds.
The half-life of the drug in chicks is about 2 days. However, it has recently been confirmed that the residual time of toltrazuril in the edible tissues of chicks is very long, and the residual drug can still be detected in the pectoral muscle 24 days after stopping the drug, therefore, the United States has not yet approved the marketing of this product by the end of the 20th century.
