Dopamine hydrochloride can be used for various types of shock, hypotension. Including toxic shock, cardiogenic shock, hemorrhagic shock, central shock, especially for patients with renal insufficiency, reduced cardiac output, increased peripheral vascular resistance, and have replenled blood volume is more meaningful. It can also treat cardiac insufficiency and acute renal insufficiency.
Uses of Dopamine hydrochloride.
Dopamine is the precursor of norepinephrine biosynthesis and one of the central transmitters, which has the effect of excitating β-receptor, A-receptor and dopamine receptor, and excitating cardiac β-receptor can increase myocardial contractility and increase cardiac output. Excitatory dopamine receptors and alpha-receptors cause dilation of the kidneys, mesentery, coronary arteries, and cerebral vessels, and increase blood flow. It has a mild contractile effect on the peripheral blood vessels and increases the arterial blood pressure. The prominent effect of this drug is to increase the renal blood flow and the glomerular filtration rate, thus promoting the increase of urine volume and urinary sodium excretion. It is clinically used for various types of shock, especially for shock accompanied by reduced cardiac contractility and renal insufficiency.
Pharmacological action of Dopamine hydrochloride.
This product is a precursor to the synthesis of epinephrine in the body, with β(mainly β1 receptor) receptor agonist, also has α receptor agonist, and can also promote the release of norepinephrine. It can enhance myocardial contractility, increase cardiac output and accelerate heart rate slightly (not as obvious as isoproterenol). Excite the α receptors of blood vessels in skin, muscle and other tissues, causing blood vessels to contract and reduce blood supply; Excitation of dopamine receptors in internal blood vessels (kidney, mesentery, heart) causes them to expand and increase blood flow. There was no significant change in total peripheral resistance, but it was beneficial to improve blood supply to vital organs during shock.
Product method of Bulk Dopamine hydrochloride.
It's made by opening a ring of piperethylamine. Add piperiethylamine and phenol to reaction pot, cool, slowly add hydrochloric acid, heat up to 110℃ reflux 12-44h. When the reaction reaches the end point, cool slightly and stir well with water. Stand stratification and separate the phenolized layer. The water layer was extracted with isopropyl acetate, and the extracted water layer was evaporated under pressure (8.0kPa) to dry, then 1/2 amount of ethanol and hydrochloric acid were added, and heated to dissolve. Cooling, crystallization, filtration, filter cake washed with ethanol, dry, to get the finished product. The yield is 74%.
