Amlodipine benzenesulfonate is a dihydropyridine calcium antagonist, which can effectively inhibit the transmembrane inward flow of calcium ions in the myocardium and vascular smooth muscle, and reduce blood pressure by directly dilating vascular smooth muscle. It has prominent effect on peripheral blood circulation and strong selectivity on blood vessels, which can diastole coronary arteries and systemic blood vessels. Increase coronary blood flow and reduce blood pressure. In vivo there is a weak negative inotropic effect on the human sinus node, atrioventricular node has no effect.
Pharmacological Effects of Amlodipine Besylate Raw Materials Powder.
Amlodipine benzenesulfonate is a dihydropyridine calcium antagonist (calcium ion antagonist or slow channel blocker). Cardiac and smooth muscle contraction is dependent on the entry of extracellular calcium ions into the cell via specific ion channels.
It selectively inhibits the transmembrane entry of calcium ions into smooth muscle cells and cardiomyocytes, with a greater effect on smooth muscle than on cardiac muscle. Its interaction with calcium channels is determined by the progressive rate of binding and dissociation to and from its receptor sites, so that pharmacological effects occur gradually.
It is a peripheral arterial dilator and acts directly on vascular smooth muscle to reduce peripheral vascular resistance, thereby lowering blood pressure. Negative inotropic effects are observed in in vitro experiments at therapeutic doses, but not in overall animal experiments. It does not affect plasma calcium concentrations.The antihypertensive effect of this product was confirmed in 15 randomised, double-blind, placebo-controlled clinical trials. Once-daily dosing in patients with mild-to-moderate hypertension reduces both prone and standing blood pressure for 24 hours, and long-term use does not cause significant changes in heart rate or plasma catecholamines. The antihypertensive effect is smooth, with little difference between peaks and troughs. The antihypertensive effect is dose-related, and the magnitude of blood pressure reduction is related to pre-treatment blood pressure. Moderate hypertensives (diastolic blood pressure 105-114 mm Hg) have a higher efficacy than mild hypertensives (diastolic blood pressure 90-104 mm Hg), and normotensives do not have a significant effect after taking the drug.
