Pharmaceutical Bulk Trospium chloride Powder Details
Trospium chloride Powder Usage and Synthesis.
Trospium chloride is a quaternary ammonium compound with anticholinergic and antispasmodic effects.
Trospium chloride acts on M receptors on the effectors innervated by cholinergic nerves and antagonises the action of acetylcholine on M receptors.
Its parasympathetic blockade induces relaxation of bladder smooth muscle, resulting in an increase in bladder capacity. At therapeutic doses, it has a weak effect on nicotinic receptors.
Uses and functions of Trospium chloride.
Trimethoprim belongs to the group of anticholinergic compounds known as muscarinic receptor antagonists, which relax the smooth muscle of the bladder and attenuate the smooth muscle contractions that lead to the symptoms of OAB. It is used as an anticholinergic agent for the treatment of overactive bladder with symptoms of urge incontinence.
Pharmacological Effects of Trospium chloride.
Trospium chloride is an artificially synthesised tropic acid derivative with four ammonium group structures, belonging to parasympathetic nerve blocking drugs, similar to atropine, acting mainly through the endogenous neurotransmitter acetylcholine competitive binding to the postsynaptic membrane of the M-receptor and the action, on the parasympathetic innervated organs play a role in reducing parasympathetic tone, removing smooth muscle spasm due to the parasympathetic nerve, and also has a certain effect on the gastrointestinal, biliary, and urinary tracts. It also has a certain effect on gastrointestinal, biliary and urinary tracts. This product has low fat solubility and does not easily pass the blood-brain barrier, and does not produce central nervous system side effects.
1. Enhance the anticholinergic effect of drugs, such as amantadine, tricyclic antidepressants, quinidine, antihistamine and verapamil; enhance the cardiac acceleration effect of sympathomimetic drugs; reduce the positive kinetic effect of drugs such as metoclopramide and cisapride.
2. Since this product may affect the dynamics and secretion of the gastrointestinal tract, it cannot be excluded that this product may affect the absorption of other drugs taken at the same time.
3. Since it cannot be excluded that drugs such as guar gum and anticholinergic amines inhibit the absorption of this product, the use of these drugs with this product is not recommended.
4. In vitro tests have shown that this product affects the metabolism of cytochrome enzymes P450 related to drug metabolism (CYP1A2, 2A6, 2C9, 2C19, 2D19, 2D6, 2E1, 3A4). No effect on other metabolism.
Production method of Bulk Trospium chloride Powder.
Tropine is acylated and hydrolysed to form nortropine, which reacts with 1,4-dichlorobutane to form (3q-nortropine)-8-spiro-1'-pyrrolidinium chloride, and then reacts with activated amide of diphenylglycolic acid to form trichlormethane under the catalysis of DMAP.
