Silodosin Usage and Function.Compared to its peers Prazosin and Tamsulosin, silodoxin is highly selective towards α1A-receptors located in the prostate and bladder neck, while its affinity for α1B- and α1D- receptors is not significant. It blocks the alpha1A-receptor at this site, relaxes smooth muscle, and leads to improved urine flow and reduced BPH symptoms.
Silodoxin binds more selectively to the α1A-receptor than to the cardiovasculary-associated α1b-receptor, maximizing activity to the target organ while minimizing potential effects on blood pressure.
Uses of Silodosin.
The α1-adrenergic receptor is a G protein-coupled receptor: by binding to its natural ligands norepinephrine and epinephrine, it activates phospholipase C and downstream signaling molecules, including inositol triphosphate and diglycerol. Eventually, intracellular calcium levels increase, so smooth muscle contracts.
Silodosin is an alpha1-adrenergic receptor antagonist with the highest selectivity for the alpha1a-adrenergic receptor subtype. By blocking the α1a-adrenergic receptor signaling pathway, silodoxin promotes relaxation of the prostate and urethral smooth muscle, thereby improving lower urinary tract symptoms such as urination. Silodoxin also targets the afferent nerves in the bladder, relieving symptoms of bladder overactivity and storage.
