Uses of Iron Sucrose.

Pharmacological Effect of Iron Sucrose.
Iron sucrose is a polynuclear iron(III) hydroxide with a core surface surrounded by a large number of non-covalently bound sucrose molecules, forming a complex with an average molecular weight of 43 kDa.
This macromolecular structure avoids being eliminated from the kidneys. This complex is structurally stable and releases iron ions under reasonable conditions.
ABSORPTION: A single intravenous dose containing 100 mg of the product results in the highest level after 10 min, averaging 538 μmol/L.
DISTRIBUTION: The volume of distribution in the central chamber is approximately 3L, and the steady-state volume of distribution is approximately 8L.
Metabolism and excretion: injected iron is rapidly eliminated from the plasma with a half-life of about 6 h. Iron clearance is less than 5% of the total clearance 4 h prior to injection of the product, and about 75% of the sucrose is excreted 24 h later.
Product Method of Bulk Iron Sucrose Powder.
(1) crystallisation: adjust the pH of sucrose solution to 1-4 with acetic acid, add iron salt solution to sucrose solution to form a mixture, adjust the pH of the mixture to 1-3 with Na2CO3 solution, stir, continue to add Na2CO3 solution to the mixture to the appearance of turbidity and the pH of the mixture is adjusted to 4-7, filter, and add water to get the discus solution;
(2) Complexation: heat the sucrose solution, add lye for alkalisation, add the discus solution to the alkalised sucrose solution, and leave it overnight to get the complexation solution;
(3) Refining: the complexing solution after standing overnight is filtered, and the filtrate is precipitated with 2-3 times of 95% ethanol and filtered, i.e. iron sucrose.
