Albendazole is a broad-spectrum thiabendazole-based anthelmintic developed by Skebichem (the predecessor of GlaxoSmithKline) for both human and veterinary use that inhibits the growth and reproduction of the worms by inhibiting the proteins in the intestinal tract or the absorptive cells of the worms, thus preventing them from ingesting the sugars on which they depend for their survival, leading to a depletion of the endogenous glycogen of the worms, as well as an inhibition of the fenvalerate reductase system, which prevents adenosine triphosphate (ATP) from being produced, resulting in a failure of the worms to survive and reproduce, and the worms will ultimately die gradually due to the depletion of their energy resources.

Albendazole Powder Usage and Synthesis.
Albendazole has the effect of completely killing the eggs of hookworms and whipworms and partially killing the eggs of roundworms, and can also kill and expel all kinds of nematodes parasitised in animals, and also has obvious killing and expelling effects on tapeworms and cysticerci.
It is suitable for the treatment of worm (encapsulation) and the nervous system infection caused by the worm in pork (cysticercosis), and is also used for the treatment of hookworm, roundworm, pinworm, trichinella, tapeworm, whipworm and faecal roundworm.
Uses of Albendazole.
1. Albendazole is a highly effective broad-spectrum anthelmintic. It can block the absorption of many kinds of nutrients and glucose by worms, leading to the depletion of the glycogen of the worms, resulting in the inability of the parasites to survive and reproduce. It is used for the treatment of single or mixed sensations caused by roundworms, pinworms, whipworms, hookworms and so on.
2. It binds microtubule protein and inhibits polymerisation of microtubules.
3. It is a new drug of high efficiency and broad-spectrum anthelmintic, with remarkable efficacy against liver flake schistosomes, tapeworms, and nematodes of lung and gastrointestinal tract. It is one of the strongest benzimidazole drugs, and it is the first choice for the prevention and treatment of parasitic diseases in livestock and poultry. The product is effective against adult and larvae of liver flake trematode of cattle and sheep, and large trematode, and the reduction rate can reach 90-100%.
In recent years, it is found that the product also has a strong effect on bovine cysticercus, cysticercus reduction after treatment, the disappearance of foci. In addition, the product can also be used to prevent parasitic infections, so as to promote the growth of sheep to increase wool production, can receive better results.
4. It is used for the prevention and control of cabbage downy mildew, rice blast, tobacco powdery mildew, anthracnose and so on.
Pharmacological Effect of Albendazole.
Albendazole is a benzimidazole derivative that is rapidly metabolised in vivo to sulfoxide, sulfonol and 2-aminosulfonol. It selectively and irreversibly inhibits glucose uptake in intestinal nematodes, depletes endogenous glycogen, and inhibits the enzyme fumaric acid reductase, preventing the production of adenosine triphosphate and causing death.
Similar to mebendazole, it causes intracellular transport blockage by inducing cytoplasmic microtubule degeneration in the intestinal cells of the worms and combining with its microtubule proteins, which leads to the accumulation of secretory particles in the Golgi apparatus, gradual dissolution of cytoplasm, and the death of the worms.
This product has the effect of completely killing hookworm eggs, roundworm eggs, pinworm eggs, trichinella eggs, tapeworm eggs and cysticercosis whipworm eggs and partially killing roundworm eggs.
Production Method of Albendazole.
(1) 2-Acetamido-4-chloronitrobenzene as raw material.
2-Acetamido-4-chloronitrobenzene was reacted with propanethiol to form 2-amino-4-propylthioaniline, and then 2-amino-4-propylthioaniline was obtained by palladium-carbon-catalytic hydrogen reduction. Finally, it was cyclised with methyl cyanoformate to obtain propylthiobenzimidazole. (2) Using o-nitroaniline as raw material.
O-nitro aniline firstly reacted with sodium thiocyanate under chlorination to form 2-nitro-4-thiocyanatobenzenamine, then reacted with bromopropane to form 2-nitro-4-propylthioaniline, which was reduced to 2-amino-4-propylthioaniline by sodium sulphide, and finally cyclohexaned with methyl cyanoamino formate to form albendazole with a total yield of 60%.
